Genomic DNA Extraction & Purification

Genomic DNA (gDNA) is the heritable, genetic information contained in each cell that provides the information needed to develop, maintain, and reproduce an organism. Organized into chromosomes, gDNA is protected by proteins and partitioned into the nucleus in eukaryotic cells. The ability to extract and purify gDNA from samples of interest is an essential first step in the study of many biological processes. Purification of gDNA often begins with homogenization of the sample followed by lysis of the cells to release the gDNA. RNase is typically employed to remove contaminating RNA from the lysate. The gDNA is then conditioned to selectively bind to a capture column, where it is subsequently washed to remove salts and other contaminants prior to elution. 

NEB offers two approaches for the extraction and purification of gDNA: the Monarch Genomic DNA Purification Kit, which is a silica spin column based process and the Monarch HMW DNA Extraction Kits, which utilize a novel glass bead based process for purification of extremely high molecular weight DNA.  

  • Monarch Genomic DNA Purification Kit

    The Monarch Genomic DNA Purification Kit (NEB #T3010) is a comprehensive kit for gDNA extraction and purification from a wide variety of sample types including blood, cells, tissues, and tough-to-lyse samples including bacteria and yeast. This kit includes lysis buffers for various input materials, Proteinase K for homogenization of some sample types, and RNase A for efficient RNA removal. The gDNA is eluted with high yield and purity and with good integrity, ready for use in downstream applications including qPCR, long range PCR, and NGS library prep.

  • Monarch HMW DNA Extraction Kits

    Available for processing cells & blood (NEB #T3050) or tissue and other sample types (NEB #T3060), the Monarch HMW DNA Extraction Kits provide a rapid and reliable process for extracting high molecular weight DNA (HMW DNA) from biological samples. Utilizing an optimized process that combines gentle cell lysis with a tunable fragment length generation followed by precipitation of the extracted DNA onto the surface of large glass beads, the prep proceeds rapidly and utilizes standard laboratory equipment. DNA size ranges from 50-250 kb for the standard protocol and into the Mb range on several sample types when the lowest agitation speeds are used. Purified DNA is recovered with high yields and with excellent purity, including nearly complete removal of RNA. Purified HMW gDNA is suitable for a variety of downstream applications including long-read sequencing (e.g., Oxford Nanopore Technologies®).
