FAQ: What buffers are recommended for shearing DNA?
DNA should be sheared in either 10 mM Tris, 1mM EDTA, pH 8.0 or 10 mM Tris, 0.1 mM EDTA, pH 8.0, since shearing in buffers other than TE causes DNA damage. However for input into the EM-seq module E7125 the DNA must be free of EDTA. Therefore a cleanup or buffer exchange is recommended between shearing and oxidation, so that the DNA is in nuclease free water.
