FAQ: What are the differences between BsmBI and its isoschizomer, Esp3I?
BsmBI is a Type IIS restriction enzyme that requires incubation at 55°C and is supplied with NEBuffer 3.1. Esp3I is an isoschizomer of BsmBI that requires incubation at 37°C and is supplied with CutSmart® Buffer (>215 restriction enzymes are supplied with CutSmart). Both these enzymes are used in Golden Gate Assembly.
