FAQ: How can I improve PmeI digestion of genomic DNA embedded in agarose?

PmeI has a difficult time cutting to completion because it looses activity before being able to penetrate the center of the agarose. Decreasing the reaction temperature  to room temperature or 4C increases survival of PmeI and allows complete cutting of the embedded genomic DNA.   For more information on how to digest agarose-embedded DNA please refer to this page in our website:https://www.neb.com/tools-and-resources/usage-guidelines/digestion-of-agarose-embedded-dna-info-for-specific-enzymes. For more information on the survival rate of our enzymes in a reaction please refer to this chart: https://www.neb.com/tools-and-resources/usage-guidelines/restriction-endonucleases-survival-in-a-reaction.
